Development
The CAG-tdTomato sequence was targeted into the Rosa26 locus to obtain the founder mice expressing red fluorescence throughout the body. The founder mice were mated with wild-type C57BL/6 mice to obtain heterozygous mice, and interbreeding of heterozygous mice resulted in homozygous mice.
This KI (knock-in) mouse line with tdTomato cDNA, under the control of the CAG promoter in Rosa26, causes all tissues except red blood cells and hair to fluoresce red under excitation light.
C57BL/6-tdTomato Mice Applications
1. In vivo cell tracking and labeling experiments.
2. Cells isolated from tdTomato red fluorescent mice may be easier to culture than primary cells from wild-type mice transfected with EGFP plasmids.
3. Tissue engineering and regenerative medicine research, which can be used to study the survival, integration, and function of transplanted cells or tissues.
