Development
The heterozygous mice with the Rosa-CAG-LSL-tdTomato-WPRE conditional allele are fertile. A loxP-flanked STOP cassette on one side prevents the transcription of the downstream red fluorescent protein variant (tdTomato). Since this CAG promoter-driven expression cassette is targeted to insert at the Gt(ROSA)26Sor locus, the expression of tdTomato depends on the tissue expressing the Cre recombinase. In the progeny of mice expressing the Cre recombinase, the STOP cassette is excised in the tissues expressing Cre, leading to the expression of tdTomato.
