Development
Linearized CAG-EGFP fragments were injected into the pronuclei of Crl:CD(SD) rat embryos to obtain transgenic positive rats. Under a fluorescence microscope, rats expressing EGFP fluorescence throughout the body were selected as founder rats. By mating the positive rats with wild-type Crl:CD(SD) rats, the SD-EGFP transgenic rat line was established.
Under the control of the CAG promoter, this transgenic rat line with the ‘enhanced’ green fluorescent protein (EGFP) cDNA causes all tissues, except red blood cells and hair, to fluoresce green under excitation light. Homozygous rats have normal viability.
SD-EGFP Rats Applications
1. Rat cell or tissue tracing
2. Cells isolated from EGFP transgenic rats may be easier to culture than wild-type rat primary cells transfected with the EGFP plasmid
