Development
The B6-Fah knockout mice are one of a series of liver injury mouse models independently developed by Vitalstar Company.
The Fah gene in mice is located on chromosome 7. The CRISPR-Cas9 technology is used to target the third and fourth exons of the Fah gene, thereby achieving gene knockout in B6 mice. The absence of the Fah gene leads to the accumulation of toxic tyrosine metabolites in hepatocytes, causing extensive and persistent liver damage. Without the control of exogenous drugs or feed, hepatocytes will gradually necrose completely, providing an opportunity for exogenous hepatocyte reconstruction in mouse livers. Mice with Fah deficiency can maintain normal survival by daily administration of NTBC drug, and liver damage can be achieved by withdrawing the drug when needed.
Therefore, B6-Fah-/- mice serve as a liver injury model.
