Development

Using random transgenic technology, the BAC containing complete hPCSK9 sequence, including introns, exons, 5'UTR and 3'UTR, were injected into fertilized mouse eggs of C57 mice. This resulted in the creation of humanized mice expressing the hPCSK9 protein. When fed a high-cholesterol diet, these mice develop hyperlipidemia, making them an ideal animal model for evaluating the efficacy of drugs targeting PCSK9 for lipid-lowering purposes.

Proprotein Convertase Subtilisin/Kexin type 9 (PCSK9) is an important regulatory factor in the body's cholesterol metabolism pathway and serves as an effective target for the development of lipid-lowering drugs. These mice can be used for research in lipid-lowering and cardiovascular diseases, particularly for evaluating small nucleic acid drugs targeting hPCSK9 (targeting 5'UTR, 3'UTR, exons, and introns).
