Development
In SD background rats, rat Rag2 and Il2rg gene-specific sgRNA (single-guide RNA) was utilized to mediate Cas9 nuclease cleavage of the genome respectively, resulting in DSBs (Double-Stranded Breaks), which were repaired by the NHEJ (Non-homologous end Joining ) repair pathway, resulting in shifted code mutations to achieve knockdown of Rag2 and Il2rg genes. This strain is a severely immunodeficient rat with no mature T cells, B cells or functional NK cells.
Compared with mice, this strain can collect much larger amounts of biological samples (e.g., tumors, blood, etc.) and is less difficult to transplant.
This strain also has tremendous advantages in terms of tumor efficacy, pharmacokinetics, and toxicology assessment.
