Development
LAG3 (Lymphocyte-activation gene 3), also known as CD223, is a cell surface protein molecule with multiple biological functions. LAG3 is expressed on activated T cells, B cells, NK cells, and plasmacytoid dendritic cells, and its main ligand is the MHC class II molecule. This protein, similar to CTLA-4 and PD-1, negatively regulates T cell proliferation, activation, and homeostasis, and has been reported to play a role in Treg suppressive function. It can also be used for LAG3 humanized antibody-related efficacy or drug screening experiments.
We used the CRISPR-Cas9 assisted method to replace the extracellular domain of mouse LAG3 with human sequences. In this way, in the humanized LAG3 mice, human LAG3 antibody inhibitors can be recognized and exert biological functions, which can be used for related efficacy or drug screening experiments
